564 Vertex and Occipital Intermediate and Terminal Hair Follicles from Androgenetic Alopecia Patients are Differentially Affected by Testosterone Ex Vivo

A key dogma of male pattern androgenetic alopecia (mpAGA) research is that clinically unaffected hair follicles (HFs) from the occipital scalp are androgen-insensitive, while frontotemporal and vertex (affected) HFs highly androgen-sensitive thus get miniaturized by testosterone (T) as consequence reduced dermal papilla (DP) inductivity increased DP fibroblast emigration. We have probed this treating organ-cultured intermediate (i) or terminal (t) affected mpAGA patients with T. Short-term treatment 30nM T tHFs indeed leads to diminished (reduced versican expression), but also promotion DC apoptosis (increased TUNEL+ caspase-3+ cells). In addition, more cells were found in treated tHFs, suggesting emigration DC. iHFs, decreased expression addition alkaline phosphatase activity DP, cell number Instead, promoted matrix keratinocyte proliferation iHFs both, i/tHFs. RNAseq analysis revealed gene changes after 24h 10nM T, incl. known androgen-regulated genes (e.g. IGF1, MMP3), both distinct transcriptomic profiles. Thus, our data provide first demonstration T-mediated effects on fibroblasts targeted “mini organs” differentially impacts i tHFs. we show no means yet display a very androgen response-profile compared same patient.
